Inactivation of Octopamine in Larval Firefly Light Organs by a High-affinity Uptake Mechanism
نویسندگان
چکیده
1. Uptake studies using radioactive octopamine have revealed that the larval lantern of the Photuris firefly possesses a concentrative, high-affinity uptake system for octopamine. This compound has been shown previously to be the neurotransmitter of the photomotor neurones. 2. Imipramine is a potent inhibitor of the uptake of octopamine in the lantern. At 10~moll~', it reduces specific uptake of radioactive octopamine with a K, of 5-8xlO~moll~. Incubation of lanterns in 2-5xlO~moll~' imipramine induces an increase in sensitivity of the luminescent response to endogenously released and exogenously applied octopamine. Release of endogenous octopamine is potentiated in imipramine-treated lanterns, which also show a significant reduction of octopamine content with this treatment. 3. Uptake of octopamine does not appear to be affected by denervation of the lanterns, suggesting that nerve terminals are not the principal sites of octopamine uptake.

Short Communication Release of Octopamine from the Photomotor Neurones of the Larval Firefly Lanterns
The biogenic amine, octopamine, has been proposed as the transmitter in the firefly light organ (Evans, 1980; Robertson, 1981). Whereas initial studies implicated adrenaline (Smalley, 1965; Carlson, 1968a), octopamine was found to be more potent in inducing luminescence than any amine other than synephrine (Carlson, 19686). Octopamine, but not synephrine, was found in adult lantern segments (Ro...
متن کاملNitric oxide and the control of firefly flashing.
Bioluminescent flashing is essential for firefly reproduction, yet the specific molecular mechanisms that control light production are not well understood. We report that light production by fireflies can be stimulated by nitric oxide (NO) gas in the presence of oxygen and that NO scavengers block bioluminescence induced by the neurotransmitter octopamine. NO synthase is robustly expressed in t...

متن کاملRole of Nitric Oxide and Mitochondria in Control of Firefly Flash1
SYNOPSIS. In light-producing cells (photocytes) of the firefly light organ, mitochondria are clustered in the cell periphery, positioned between the tracheolar air supply and the oxygen-requiring bioluminescent reactants which are sequestered in more centrally-localized peroxisomes. This relative positioning suggests that mitochondria could control oxygen availability for the light reaction. We...
متن کاملMathematical Model of the Firefly Luciferase Complementation Assay Reveals a Non-Linear Relationship between the Detected Luminescence and the Affinity of the Protein Pair Being Analyzed.
The firefly luciferase complementation assay is widely used as a bioluminescent reporter technology to detect protein-protein interactions in vitro, in cellulo, and in vivo. Upon the interaction of a protein pair, complemented firefly luciferase emits light through the adenylation and oxidation of its substrate, luciferin. Although it has been suggested that kinetics of light production in the ...
